FITC-derivatives

The labelling of dextran with fluorescein via its derivative fluorescein isothiocyanate (FITC) was first described in a publication in 1973 by de Belder and Granath (ref.1). Now it is a long-established method for obtaining fluorescent-labelled polysaccharides. These products are designated as FITC-derivatives. The fluorescein moiety is bound by a thiocarbamoyl-linkage which displays good stability in-vitro and in-vivo.

TdB labs produce a wide variety of FITC-derivatives with a mean molecular weight ranging from 4 kDa to 2000 kDa. All our FITC products are supplied as a yellow to orange powder.

Spectral data

Maximum fluorescence is recorded in the pH range 6-8.

Excitation maximum: 490 - 495 nm.
Emission maximum: 520 ± 5 nm.

Charged derivatives
Our standard range of FITC products also comprises derivatives with functional groups. This includes carboxymethyl (CM)-derivatives which display a negative charge (anionic). Our diethylaminoethyl (DEAE) derivatives are positively charged (cationic). We have also developed a derivative called Q, which is a substituent of O-Trimethylammonium-glycidyl.

Fixable derivatives
FITC-lysine derivatives are polysaccharides carrying the natural amino acid Lysine as a substituent.

Applications
FITC-labelled polysaccharides have been used for more than 30 years, largely for real-time studies of permeability, microcirculation, distribution and diffusion.

Derivatives with diethylaminoethyl (DEAE)-groups or carboxymethyl (CM) are often used to study the effects of charge on permeability

Lysine modification provides tools for fixation and conjugation in order to preserve components in a “life-like” state. Read more about applications here.

FITC-dextran

FITC-dextran is produced by FITC-dextrans are synthesised by coupling fluoresceinyl isothiocyanate (5-isomer) to selected dextran fractions. TdB Labs produce FITC-dextrans from 4 kDa to 2000 kDa.

FITC-lysine-dextran

FITC-lysine-dextran is a derivative carrying both free primary amines and free carboxylates as well as the fluorescent dye Fluroescein (FITC). TdB Labs with a molecular weight from 4 kDa to 500 kDa.

FITC-CM-dextran

FITC-CM-dextrans are standard FITC-dextran carrying carboxymethyl-substituents. TdB Labs produce FITC-CM-dextrans from 4 kDa to 150 kDa.

FITC-CM-polysucrose

FITC-CM-polysucroses are standard FITC-polysucrose carrying carboxymethyl-substituents. TdB Labs produce FITC-CM-polysucroses with a molecular weight of 70 kDa and 400 kDa.

FITC-DEAE-dextran

FITC-DEAE-dextran are produced by introducing cationic diethylaminoethyl groups into standard FITC-dextran. TdB Labs produce FITC-DEAE-dextrans from 4 kDa to 150 kDa.

FITC-DEAE-polysucrose

FITC-DEAE-polysucroses are standard FITC-polysucroses carrying cationic (positively charged) diethylaminoethyl groups. TdB Labs produce FITC-DEAE-polysucrose with the molecular weight of either70 kDa or 400 kDa.

FITC-dextran sulfate

FITC-dextran sulfate is dextran sulfate sodium with a mean molecular weight of 40 kDa labelled with FITC.

FITC-hydroxyethyl starch

FITC-HES (Fluorescein isothiocyanate hydroxyethyl starch) is manufactured by reacting FITC with HES. The mean average molecular weight is approx. 165 kDa.

FITC-inulin

FITC-inulin is synthesised by reacting fluoresceinyl isothiocyanate (FITC) with inulin from dahlia tubers. TdB Labs produce FITC-inulin with a molecular weight of 4 kDa.

FITC-polysucrose

FITC-Polysucrose is prepared by reacting polysucrose with FITC. TdB Labs produce FITC-polysucrose from 20 kDa to 400 kDa.

FITC-Q-dextran

FITC-Q-dextran is a positively charged polysaccharide labelled with FITC. TdB Labs produces a FITC-Q-dextran with a molecular weight of 10 kDa.

FITC-trehalose

FITC-Trehalose (Fluorescein isothiocyanate trehalose) is a fluorescent derivative of trehalose with molecular weight of 731.7 Da.

Fluorescein Hyaluronic Acid

Hyaluronic acid (HA) can be obtained from various sources, for example rooster combs or Streptococcus equi. We can supply derivatives of either of these qualities labelled with fluorescein (F-Ha).
